چکیده

(1) Background: Temozolomide (TMZ), an oral alkylating agent, is used to treat malignant gliomas and other difficult-to-treat tumors. TMZ can enter the cerebrospinal fluid p.o. (per os) does not need hepatic metabolism for activation of its use as a standard chemotherapeutic regimen after surgical resection glioma brain. However, prognosis remains poor most patients, survival rate still unsatisfactory. Gallic acid (Ga) secondary metabolite existent in numerous plants. Ga shows various bioactivities, including antioxidant, anti-inflammatory, anticancer antimicrobial effects. In this study, latent enhanced anti-cancer efficacy TMZ-treated U87MG cells (a human line) was evaluated. (2) Methods: The cell line cultured 24 h. were incubated with alone, or their combination time points. Cell viability drug index evaluated by XTT-based analysis isobologram analysis, respectively. DNA destruction intracellular reactive oxygen species (ROS) generation analyzed flow cytometer. expression proteins assessed via Western blotting. (3) Results: Compared action alone TMZ/Ga augmented inhibition cellular apoptotic level line. This effect correlated decreased Bcl-2 p-Akt, corresponded p38 mitogen-activated protein kinase (MAPK) pathway. addition, suppressed TMZ-promoted ROS generation. (4) Conclusions: augment repression Akt enhancement MAPK Our results offer novel probable approach medical treatment glioma.

p38 MAPK and PI3K/AKT signalling cascades in Parkinson’s disease

Parkinson's disease (PD) is a chronic neurodegenerative condition which has the second largest incidence rate among all other neurodegenerative disorders barring Alzheimer's disease (AD). Currently there is no cure and researchers continue to probe the therapeutic prospect in cell cultures and animal models of PD. Out of several factors contributing to PD prognosis, the role of p38 MAPKs (mitog...

Evaluation of Anti-cancer activity of Boswellic acid and Montelukast sodium against human prostate cancer cell line PC-3

Prostate cancer is a devastating disease for which current therapies are inadequate. Various lines of evidences have suggested the 5-lipoxygenase (5-LOX) pathway and the leukotriene receptor pathway are potential targets for prevention or treatment of Prostate cancer. Thus, search for new anti-cancer drugs targeting 5-LOX and leukotriene is very essential and important. The objective of the pre...

Thymoquinone synergistically potentiates temozolomide cytotoxicity through the inhibition of autophagy in U87MG cell line

Objective(s): Glioblastoma multiforme (GBM) is one of the most lethal forms of human cancer and temozolomide (TMZ) is currently part of the standard treatment for this disease. Combination therapy using natural substances can enhance the anti-cancer activity of TMZ. The purpose of this study was to evaluate the effect of TMZ in combination with thymoquinone (TQ) on human GBM cell line (U87MG). ...
